This MaxSignal Total Deoxynivalenol Immunoaffinity Column can extract Deoxynivalenol (Vomitoxin, DON), as well as its two prominent acetylated derivatives, 3-acetyldeoxynivalenol (3-ADON) and 15-acetyldeoxynivalenol (15-ADON), which has a highly targeted purification effect. The purified sample solution can be directly used for HPLC or LC-MS/MS analysis after concentrating with nitrogen gas.
DON belongs to the single-endomycin family of molds and is produced by certain Fusarium fungi. 3-ADON and 15-ADON are derivatives of DON. These toxins are mostly distributed in grain seeds such as wheat, barley, and corn. Due to their high cytotoxicities and immunosuppressive properties, they pose a health threat to humans and animals.
The basis of the IAC measurement is the antigen-antibody interaction. All main toxin antibodies are embedded throughout the column. After a sample is extracted and filtered, it is slowly passed through the IAC. The toxins bind to their corresponding antibody in the column. The IAC is then washed to remove unbound substances. The toxins are then eluted with the eluent, concentrated using nitrogen gas, then injected into an analytical instrument for detection. Affinity columns can be used in combination with HPLC and LC-MSMS to achieve rapid testing, and to increase signal-to-noise ratio and improve the accuracy of the detection method.
